443 Inducible IL-2Rγ/IL-4Rα receptor in keratinocytes is involved in epidermal barrier alterations

Atopic dermatitis (AD) is an inflammatory skin disease triggered by a vicious circle between epidermal barrier alteration and immune system dysregulation. This can notably be initiated exposure of epidermis to interleukins (IL) 4 13 that alter keratinocyte phenotype through activation their IL-4Rα/IL-13Rα1 (type II) receptor. Expression IL-2Rγ/IL-4Rα I) receptor for IL-4 normally limited hematopoietic cells, however, gene expression analysis in reconstructed human (RHE) exposed IL-4, IL-13 IL-25 revealed IL-2Rγ being expressed keratinocytes. To investigate if the induced type I could AD keratinocytes, IL-2Rγ-encoding mRNA protein was firstly confirmed RHE IL-13, as well made N/TERT keratinocytes (N/TERT-RHE). Keratinocyte partially inhibited incubation with JAK inhibitors, totally IL13RA1 KO N/TERT-RHE. Consequently, when activated STAT6 MAPK signaling pathways result from both receptors. determine activate cell RHE, II receptors blocked anti-IL-13Rα1 antibodies.
